Redox Biology in Thrombosis

Description

Reactive oxygen species (ROS) and redox biology has received significant attention in vascular biology. However, ROS production by, and influences on, platelet function has received less attention than its interactions with other cells in the vasculature. Platelets both produce and respond to ROS. This session will discuss how ROS signaling regulates platelet function in ischemic pathologies. In addition, the use of ROS scavengers will be explored as a therapeutic strategy to ameliorate platelet destruction.

Dr. Craig Morrell will focus on mechanisms of platelet signaling in response to endogenous and exogenous ROS.  Furthermore, he will discuss how ROS changes the platelet proteome independent of the megakaryocyte. These data will be placed in the context of ischemic vascular pathologies, including peripheral artery disease.

Dr. Karen Vanhoorelbeke will discuss the efficacy of N-acetyl cysteine (NAC) in the treatment of immune mediated thrombotic thrombocytopenia purpura (TTP) as tested in animal models. In addition, literature and clinical trials where NAC has been used to treat TTP patients will be reviewed. Finally, novel data on the role of anti-ADAMTS13 autoantibodies in immune-mediated TTP will be discussed.
